You do need to run all of your spyware killers in safe mode.  Make sure 
they're all updated first, then reboot and hold down F8 and pick 'safe 
mode" when the menu comes up.

Depending on what you've picked up, it can be easy to remove or it can 
be a complete bastard.  A lot of the browser hijacks hide system files 
with random names as well as system processes that know how to hide, and 
also know how to rename themselves when you shut down normally so they 
can reappear on bootup and reinstall the files you deleted the first time.

Some of them just take a good whacking by someone experience in what to 
look for.  I've had to clean out some machines that actually took me 6 
hours of meticulous, highly frustrating hunting, searching, and 
downloading to clean out.  If the customer didn't have non-backed-up 
stuff on the machine, it would have been simpler and cheaper for me to 
wipe the SOB and reinstall everything.

Browser hijacks are usuall a complete bear to kill.  You have to clean 
out not only system files, but also registry entries.  And you have to 
make sure you kill and remove running processes that will take hidden 
files and reinstall themselves from them.

If your hijack has any specific traits, you can google for some 
remedies.  A program called Hijack This is very good for finding 
nastiness, but it's not a tool for the faint-hearted or inexperienced, 
since it allows you to remove a whole bunch of stuff that may look 
questionable, but if you don't know enough to know what's good and what 
is bad, you can render your machine unbootable.
